神經(jīng)軸突退化變性壞死是急性神經(jīng)損傷如脊髓損傷和腦創(chuàng)傷以及很多與衰老相關的慢性神經(jīng)退行性疾病如老年癡呆癥(AD)、帕金森氏癥(PD)、亨廷頓氏癥(HD)、多發(fā)性硬化癥(MS)、肌萎縮性脊髓側(cè)索硬化癥(ALS)等疾病中的常見病變。此外,糖尿病、青光眼和化療引起的神經(jīng)毒性也會導致繼發(fā)性的神經(jīng)軸突的退化變性壞死。由于人類的中樞神經(jīng)在受損或變性壞死后再生能力極其有限,如何保護受損或患病的神經(jīng),預防、延緩、阻止乃至逆轉(zhuǎn)神經(jīng)軸突退化變性壞死是生物醫(yī)學領域的一個重大前沿課題。
方燕姍課題組致力于急性神經(jīng)損傷和慢性退行性疾病中神經(jīng)退化的細胞和分子機理的研究,運用全新的神經(jīng)損傷活體動物模型結合分子遺傳學、細胞生物學、生物化學、藥物化學、超高分辨率熒光成像、新一代測序技術、現(xiàn)代蛋白質(zhì)組學和代謝組學等技術研究調(diào)控神經(jīng)退化和促進神經(jīng)再生修復的分子機制,并以此作為基礎開發(fā)用于疾病診斷治療的生物標志物和新型藥物。
